摘要
目的:用正交试验法考察甘木通总黄酮的最佳提取条件。方法:以芦丁为对照品建立标准曲线方程用于测定甘木通总黄酮含量,以乙醇为溶剂分别考察温度、乙醇浓度、提取时间、药材/溶剂比例及提取次数等因素对甘木通总黄酮提取率的影响,据此设计四因素三水平正交试验,系统考察甘木通总黄酮的最佳提取条件。结果与结论:药材与70%乙醇按照1:20的比例,在90℃回流提取1h共2次,甘木通总黄酮提取率可达93.27%。
Objective:To study the optimum technology of extracting total flavoniods from Clematis filamentosa Dunn by orthogonal test.Methods:Taking rutin as a comparison to make standard curve to determinate the content of total flavoniods.The factors that affect extracting rate of total flavoniods in Clematis filamentosa Dunn was studied,including the extracting time,temperature,ethanol concentration,ratio of solid-to-liquid and number of extraction times,and an orthogonal test table L9(3 4 )was used to investigate the best extracting condition.Results and Conclusion: Results showed that 70% ethanol as extracting solvent, ratio of solid-to-liquid 1: 20, temperature 90℃; and extracting time of lh were optimal.The extracting rate of total flavoniods from Clematis filamentosa Dunn could achive 93.27% by extracting twice under the optimum condition.
