低压缺氧对大鼠全脑缺血/再灌注后海马CA1区GLT-1表达的影响 被引量:2

摘要 目的探讨低压缺氧预处理对大鼠全脑缺血/再灌注后的神经保护作用。方法将Wistar大鼠随机分为假手术组、低压缺氧对照组、全脑缺血/再灌注组、低压缺氧预处理+全脑缺血/再灌注组。间断暴露于低压缺氧(大气压为70.66~70.93kp)环境的大鼠[(1次/d)×4d]作为低压缺氧对照组。采用Pulsinelli四血管闭塞法复制大鼠全脑缺血/再灌注模型,夹闭颈总动脉造成全脑缺血8min后再灌注。硫堇染色观察大鼠海马组织学改变;免疫组织化学方法观察神经胶质细胞谷氨酸转运体亚型-1阳性细胞表达变化,并将四组结果进行比较。结果Wistar大鼠低压缺氧预处理全脑缺血/再灌注组海马CA1区存活细胞数较全脑缺血/再灌注组明显增加,其神经胶质细胞谷氨酸转运体亚型-1阳性细胞表达量较全脑缺血/再灌注组显著升高。结论低压缺氧可通过上调神经胶质细胞谷氨酸转运体亚型-1的表达,减少神经细胞的死亡,发挥神经保护作用。 Objective To study the neural protection of Hypobaric Hypoxia Preconditioning (HHP) after global cerebral ischemia and reperfusion. Methods The Wistar rats were randomly divided into sham group, HHP control group,global cerebral ischemia and reperfusion group(IR) and HHP + IR group. HHP control group was explored environment with intermittent ( 70.66 - 70.93 kp, 1 time/d ·4d). Global cerebral ischemia was performed by four vessels occlusion in rats. We observed the changes of hippocampus neurons with thionin staining and expression of GLT-1 on hippocampal CAI subfield with immunohistochemistry staining , then the data analyzed with SPSS software. Results The quantity of the surviving cells in CA1 region of hippocampus increased in the group of HHP + IR than that of IR group , the expression of GLT - 1 on the CA1 subfield hippocampus significantly increased compared with IR group. Conclusion Hypobaric Hypoxia Preconditioning might selectively increase the expression of GLT - 1 and reduce the number of death cells of global cerebral ischemia and reperfusion . It is considered that HHP plays an important role in the neural protective effect.
