TGF-β_1影响人牙周膜成纤维细胞分泌基质金属蛋白酶-1的初步研究 被引量:3

摘要 目的:研究转化生长因子β1(transforming growth factor-β1,TGF-β1)在体外对人牙周膜成纤维细胞(human peri-odontal ligament fibroblasts,HPDLFs)分泌基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)的影响,进而探讨TGF-β1对HPDLFs分泌MMP-1的调控作用,以期为通过生物学途径建立防治正畸过程中牙根吸收的有效方法提供理论依据。方法:原代培养HPDLFs,取传代至第5代HPDLFs实验。设立TGF-β1工作浓度梯度:0.03ng/ml、0.05ng/ml、0.07ng/ml、0.09ng/ml;0ng/mlTGF-β1为空白对照组;选取4h、8h、12h、24h、48h5个时间点收获上清液。双抗体夹心酶联免疫吸附实验(enzyme-linked-immunosorbent serologic assay,ELISA)法检测各样本中MMP-1含量。采用多元线性回归方法分析实验结果。结果:MMP-1分泌量与TGF-β1作用时间总体上存在时间依赖性,具有统计学差异,P<0.05。但各作用时间下MMP-1分泌量与TGF-β1工作浓度并没有表现出一致的剂量依赖关系;4h时各实验组MMP-1分泌量随TGF-β1浓度的递增而下降;8h、12h、24h时,各实验组MMP-1分泌量与TGF-β1工作浓度基本呈正相关;48h时各组MMP-1含量变化较大没有明显趋势。结论:MMP-1分泌量与TGF-β1作用时间存在显著时间依赖性,TGF-β1可通过促进MMP-1分泌参与正畸过程中牙根吸收的发生和发展。 Objective To investigate the biological effects of transforming growth factor-β1 (TGF-β1) on human periodontal ligament fibroblasts (HPDLFs) secreting matrix metalloproteinase-1 (MMP-1). Methods HPDLFs were exposed to TGF-β1 at various concentrations (0.03ng/ml,0.05ng/ml,0.07ng/ml and 0.09ng/ml) in monolayer cultures.The levels of MMP-1 secreted by the TGF-β1-treated HPDLFs were measured using an enzyme-linked immunosorbent assay.Differences between experimental and contol groups were tested using multiple linear regression analysis. Results Our data demonstrate TGF-β1 treatment significantly increased the secretion of MMP-1 in a time-dependent manner(P〈0.05). However,differently concentrated TGF-β1 effect HPDLFs secreting MMP-1 differently at each moment. Conclusion The secretion of MMP-1 is regulated by TGF-β1. TGF-β1 play an important role in root resorption during orthodontic tooth movement.
