茶氨酸体外对人胃癌细胞MGC803增殖的作用及与阿霉素的联合作用 被引量:6

EFFECT OF THEANINE OR COMBINED WITH DOXORUBICIN ON PROLIFERATION OF HUMAN GASTRIC CARCINOMA CELL LINE MGC803 IN VITRO

摘要 目的研究茶氨酸(theanine)对人胃腺癌细胞株MGC803增殖的影响及其与阿霉素(doxorubicin,DOX)的联合作用。方法采用MTT法结合形态学观察,研究茶氨酸单独或与DOX联合作用对MGC803增殖活性的影响;采用荧光分光光度法检测茶氨酸对MGC803细胞内DOX药物浓度的影响,探讨茶氨酸对DOX化疗敏感性的影响。结果2.0、4.0μmol/L茶氨酸单独作用于MGC803后其吸光度值与空白对照组相比有统计学差异(P<0.01);茶氨酸与DOX联合作用后对MGC803增殖的抑制率明显提高,并观察到典型的凋亡细胞形态学特征;DOX联合茶氨酸作用后,使MGC803细胞内DOX浓度升高1.35倍。结论茶氨酸可能有直接抑制MGC803细胞增殖的作用,并使肿瘤细胞内DOX滞留,提高了肿瘤细胞对DOX的化疗敏感性。 Objective To study the effect of theanine or combined with doxorubicin (DOX) on proliferation of human gastric cancer cell line MGC803. Method Using tetrazolium microculture (MTT) assay combined with morphological observation, we observed the effects of theanine with or without DOX on proliferation activity of MGC803. The impact of theanine on DOX influx and efflux in MGC803 cells were detected by fluorescence spectrography, for approaching the effect of theanine on chemotherapeutic drug sensitivity of MGC803. Results MTT assay showed statistical difference of absorbance values at 2.0 lamol/L and 4.01amol/L concentration of theanine as compared with untreated controls (P〈0.01). The inhibition ratio was obviously improved by DOX combined with theanine. The typical cyto-morphology of apoptosis cells was also observed. The results of fluorescence spectrography showed that theanine increased the intracellular concentration of DOX in tumor cell by 1.35-fold. Conclusion Theanine may directly inhibit the proliferation of gastric carcinoma cell and inhibit the efflux of DOX, then to enhance its chemotherapeutic drug sensitivity to tumor cell.
