摘要
目的:观察巨噬细胞(macrophage,MΦ)对肝癌细胞株的凋谢作用。方法:从肝癌患者腹水中分离MΦ,以细菌脂多糖(lipopolysaccharides,LPS)进行激活,人肝癌细胞株SMMC7721为靶细胞,光镜观察靶细胞形态学改变。结果:LPS活化的MΦ与靶细胞共同培养1h可见紧密接触,24h靶细胞呈现核碎裂,胞浆嗜酸性增强,凋谢小体形成等变化;未活化的MΦ仅有与靶细胞接触但未见其破坏现象。结论:活化的MΦ通过凋谢的方式引起肝癌细胞株死亡。
Objective: To observe apoptotic phenomena of hepatic carcinoma cell lines induced by macrophages.Methods: The peritoneal macrophages were obtained by collecting ascites in patients with hepatocellular carcinoma and activated by lipopolysaccharides.SMMC 7721 ,a human hepatic carcinoma cell line,was served as target cell.Morphologic changes of the target cells were distinguished with light microscopy.Results: After macrophages and target cells cultured together,activated macrophages closely integrated with the target cells in 1 h,and the target cells appeared in the karyorrhexis,cytoplasm acidophil enhancement,apoptotic bodies,and so on in 24 h.No-activated macrophages could not distroy the target cells in 24 h Conclusion: Activated macrophages mediate hepatic carcinoma cell lines death with apoptosis.
