摘要
目的:制备邻苯二甲酸酐(phthalicanhydride,PA)大鼠哮喘模型,探讨细胞间粘附分子1(intercelularadhesionmolecule1,ICAM1)与PA哮喘气道炎症的关系。方法:用PA免疫大鼠制备哮喘模型,观察哮喘大鼠支气管肺泡灌洗液中炎性细胞的变化及支气管肺组织的病理改变;用Northern杂交方法检测哮喘大鼠肺组织ICAM1mRNA的表达。结果:哮喘大鼠支气管肺泡灌洗液中炎性细胞数较对照组显著增加,支气管肺组织炎性细胞浸润程度较对照组增加,支气管粘膜皱缩加重;哮喘大鼠肺组织ICAM1mRNA比对照组表达增高,表达量与炎性细胞的改变呈显著正相关。结论:哮喘大鼠肺组织ICAM1mRNA表达量的增高不仅与哮喘气道炎症的发生有关。
Objective: To develope the asthmatic rat model induced by phthalic anhydride (PA) and the relationship between intercellular adhesion molecule 1 (ICAM 1) and to explore the airway inflammation of PA induced asthma. Methods: (1)The rat model of asthma was developed by immunization with PA to observe the changes of different inflammatory cells in the bronchoalveolar lavage fluid and the pathologic changes in the bronchus of the rat model. (2)The expression of ICAM 1 mRNA in the lung of the model was investigated with Northern blotting method. Results: The significant increase of the inflammatory cells in the bronchoalveolar lavage fluid, the increased infiltration of the inflammatory cells at the base of the bronchial mucosa and the increased folding of the mucosa were observed in PA induced asthmatic rat model. The enhanced expression level of ICAM 1mRNA was also found in this model, and the expression level was parallel with the changes of the inflammatory cells. Conclusion: The expression of ICAM 1mRNA in the lung of asthmatic rats not only participated in the development of the airway inflammation, but also closely related to the infiltration level of the inflammatory cells.
