摘要
研究壮骨颗粒含药血清对骨髓基质干细胞体外成骨活性的影响。方法:采用体外细胞培养技术,采用壮骨颗粒对细胞进行体外诱导培养,并设置空白血清对照组,进行MTT、免疫组织化学染色及ALP测定,观察壮骨颗粒对骨髓基质细胞的诱导作用。结果:在促细胞增殖方面:实验组作用显著(P<0.01),在第3d开始增殖,第6d达到平台期;而对照组对数增殖期明显延长(P<0.01);在促细胞分化方面:壮骨颗粒血清组在2d、3d、5dALP活性水平高于对照组(P<0.01)。常规形态学观察,壮骨颗粒组诱导的细胞在形态上类似于成骨细胞,免疫组化染色提示诱导细胞具有骨细胞表型。结论:壮骨颗粒组应用可促进细胞增殖,并诱导骨髓基质细胞向成骨细胞分化。
Objective: To investigate effect of Zhuanggu Granula medicated serum on proliteration and differentiation of bone marrow stromal cells(BMSCs) in vitro. Methods: Isolated BMSCs were cultured in vitro. The cells were treated with Zhuanggu Granula medicated serum, and the blank serum as the control group. The proliferation of ceils was detected by MTT assay, the macroscopic histology, HE staining and immunohistoc-hemical examinations. Results: Zhuanggu Granula medicated serum effectively stimulated BMSCs proliferation (P〈0.01). On the effect in promoting differentiation, the ceils induced by Zhuanggu Granula medicated serum was similar to osteoblasts in morphology, and immunohistochemical examinations showed the cells possessed phenotype of osteoblasts. Conclusion: Zhuanggu Granula can induce the BMSCs proliferation, and effectively stimulate BMSCs differentiating into the osteoblasts.
