摘要
目的 观察转移性不同的3种膀胱癌细胞株中Raf激酶抑制蛋白(RKIP)的表达,探讨其与膀胱癌侵袭、转移的关系.方法 选取膀胱移行细胞癌细胞株EJ、T24和BIU-87,用Western blot检测各细胞株中RKIP的蛋白表达,用逆转录-聚合酶链反应(RT-PCR)检测各系RKIP的mRNA水平,比较它们的差异.结果 EJ、124和BIU-87三者RKIP蛋白相对表达量分别为1.99±0.24、0.82±0.16、0.15±0.03(P〈0.05),mRNA相对表达量分别为1.87±0.33、0.86±0.25、0.29±0.12(P〈0.05),均依次降低.结论 RKIP可抑制膀胱癌细胞株的侵袭、转移,其表达下调可能在膀胱癌的进展中具有重要作用,RKIP可能与膀胱癌的侵袭和转移呈负相关.
Objective To evaluate the relationship between Raf kinase inhibitor protein(RKIP) and bladder cancer metastasis.Methods Thlee kinds of bladder cancer cell lines with different metastatic potentials (transitional cell carcinoma cell lines EJ,T24 and BIU-87)were observed.RKIP protein and mRNA expression was detected with Western blot and RT-PCR,respectively.Differences between the protein and mRNA expressions in each cell line were compared.Results Under normal culture condition,the relative expression of RKIP protein in each cell line(EJ,T24 and BIU-87)was 1.99±0.24,0.82±0.16,and 0.15±0.03(P〈0.05),and that of RKIP mRNA was 1.87±0.33,0.86±0.25,and 0.29±0.12 respectively.They were reduced in sequence and there Was significant difference among the three groups(P〈0.05).Conclusion RKIP may inhibit the metastasis and invasion of bladder cancer,and its down-regulation may play an important role in bladder cancer progression.
