摘要
目的:研究黄芪药材中黄芪皂苷的提取工艺及含量测定。方法:采用70%乙醇回流提取,水饱和正丁醇萃取,D101大孔吸附树脂富集纯化黄芪总皂苷,硅胶柱层析分离不同的皂苷组分;采用薄层色谱法(TLC)、高效液相-示差检测器法(HPLC-RID)进行皂苷类成分的鉴定和含量测定。结果:从黄芪总皂苷中分离出六种黄芪皂苷化合物(AS-Ⅰ~AS-Ⅵ),其中AS-Ⅰ部分经鉴定主要含皂苷Ⅱ,质量分数为29.4%,AS-Ⅲ部分经鉴定主要含皂苷Ⅳ,质量分数为22.82%。其余4个部分有待于进一步鉴定。结论:此法能较好的分离和纯化黄芪皂苷,TLC和HPLC-RID法操作简便,可作为黄芪皂苷的鉴定和含量测定方法。
Objective:Study on extracting technology and assay of astragaloside in Astragalus.Method: 70% ethanol circumfluence extraction was used.Total astragaloside was enriched and purified by extraction refine through water saturated of n-butanol and D101 mcroporous adsorption resin.Different components of astragaloside were isolated by silica gel laminar analysis.Identification by using TLC method and determination the contents by u-sing HPLC-RID Method.Result: Six compounds were isolated from total astragaloside(AS-Ⅰ ~ AS-Ⅵ),and 29.4% astragaloside Ⅱ was identified in As-Ⅰ part.AS-Ⅲ which content is 22.82% is mainly contained Astraga-losideⅣ by identification.The rest four compounds are required to be identified.Conclusion:The method can be used for astragaloside,HPLC-RID method can be used to determine the content of astragaloside.
