摘要
目的探讨糖皮质激素对哮喘小鼠肺组织水通道蛋白5(AQP5)表达的影响。方法45只清洁级BALB/c小鼠随机分为对照组、哮喘组和地塞米松干预组,每组15只。采用免疫组化法检测肺组织中AQP5蛋白的表达。结果哮喘组肺组织AQP5蛋白表达较对照组明显减低[(0.1186±0.0007)vs.(0.2060±0.0005)](P<0.01),地塞米松干预后AQP5蛋白表达较哮喘组增加[(0.1502±0.0008)vs.(0.1186±0.0007)](P<0.01)。结论哮喘小鼠肺组织AQP5蛋白表达下调,AQP5可能参与了哮喘发病时气道黏液分泌的调节,糖皮质激素可上调哮喘小鼠肺组织AQP5蛋白的表达。
Objective To study the effects of glucocorticoid on the expression of aquaporin-5(AQP5)protein in asthmatic mice lung.Methods Fourty-five pathogen-free BALB/c mice were randomly divided into three groups of control,asthmatic and dexamethasone interference with 15 mice each.The expression of AQP5 protein in the lungs was detected by immunohistochemical assay.ResultsThe expression of AQP5 protein in asthmatic group was less than that in control group[(0.1186±0.0007)vs.(0.2060±0.0005)](P0.01),which was increased in dexamethasone interference group[(0.1502±0.0008)vs.(0.1186±0.0007)](P0.01).Conclusion The expression of AQP5 protein is downregulated in asthmatic mice lung.AQP5 may be involved in airway mucus secretion of asthma.Glucocorticoid can upregulate the expression of AQP5 protein in asthmatic mice lung.
