PEDF对大鼠角膜碱烧伤后新生血管生长抑制作用的实验研究

摘要 目的 探讨色素上皮细胞衍生因子(PEDF)对大鼠角膜新生血管形成及血管内皮生长因子(VEGF)表达的影响.方法 大鼠随机分为3组:PEDF治疗组、生理盐水对照组、正常组.建立碱烧伤诱导的大鼠角膜新生血管的动物模型,治疗组和对照组分别给予l5nmol/L PEDF滴眼液和生理盐水点眼,采用裂隙灯观察新生血管的生长情况,用免疫组织化学方法检测VEGF的表达.结果 各时间点新生血管的面积,治疗组均低于对照组(P〈0.05).免疫组化显示,碱烧伤后7,10,14d治疗组VEGF的表达较对照组明显减少(P〈0.05).结论 PEDF通过下调VEGF的表达,最终可有效抑制碱烧伤诱导的大鼠角膜新生血管形成. Objective To investigate the depressant effect of pigment epithelium-derived factor (PEDF) on neovascularization and as well as the influence on the expression of vascular endothelial growth factor( VEGF ) in corneal alkali burn of rat model. Methods The rats were randomly divided into three groups : the treatment group ( received 15nmol/L PEDF drops) , the control group( treated with physiologic saline) and normal group. Corneal neovessels( CNV) model was established by alkaline burning. The growth of neovessels was observed under slit lamp microscopy. The expression of VEGF in CNV of each rat was detected by immunohistochemical staining. Results Compared with the control group, the growth of CNV in treatment group on each time point reduced obviously ( P 〈 0.05 ). The expression level of VEGF in treatment group was lower than that in the control group 7,10 and 14 days after alkali-burn. Conclusion PEDF can efficiently inhibit the growth of CNV induced by alkaline burn through reducing the expression of VEGF in cornea.
