液氮冻融洗涤血小板对人牙髓细胞增殖的影响 被引量:8

摘要 目的:与牛凝血酶激活富血小板血浆方式比较,观察液氮冻融洗涤血小板促进人牙髓细胞增殖的效果。方法:取健康志愿者因正畸需要拔除的牙齿,原代培养牙髓细胞,传代培养至第4代后分别加入不同浓度冻融激活的洗涤血小板和牛凝血酶激活的富血小板血浆,细胞定量测定试剂盒测定细胞增殖情况。结果:10~100mL/L的洗涤血小板明显促进了人牙髓细胞的增殖,而且该作用明显优于相同浓度的牛凝血酶激活的富血小板血浆。200mL/L的洗涤血小板和富血小板血浆促进人牙髓细胞增殖的作用均较100mL/L浓度组明显降低。结论:液氮冻融激活的洗涤血小板、牛凝血酶激活的富血小板血浆在一定浓度下均可以明显促进人牙髓细胞的增殖,而且前者的作用效果较后者更好。 AIM: The effect of washed platelet activated by liquid nitrogen freezing-thawing on the proliferation of human dental pulp ceils was compared with that of bovine thrombin activated platelet-rich plasma. METHODS: Human dental pulp cells from healthy extracted teeth donated by patients under orthodontic treatment were cul- tured and passaged. Different concentrations of freezing-thawing activated washed platelets or bovine thrombin - activa- ted platelet-rich plasma were added to passage 4 human dental pulp cells, respectively. The proliferation of human dental pulp cells were determined by cell quantitative measurement kit. RESULTS: Activated washed platelet in con- centration 10 mL/L to 100 mL/L evidently promoted the proliferation of human dental pulp cells. The effect was stron- ger than that of bovine thrombin-activated platelet-rich plasma in the same concentration. The effects of promotion were much lower when they were in 200 mL/L respectively. CONCLUSION: Both liquid nitrogen freezing-thawing activated washed platelet and bovine thrombin-aetivated platelet-rich plasma can promote the proliferation of human dental pulp cells, and the former shows stronger stimulatory effects on human dental pulp proliferation than the latter.
