芪苈强心胶囊活性部位中的组成成分研究 被引量:20

摘要 目的分离鉴定芪苈强心胶囊活性部位中的成分。方法采用溶剂提取和大孔吸附树脂柱色谱的方法分离芪苈强心胶囊的不同部位,通过活性筛选确定有效部位,利用HPLC-PDA/ELSD进行活性部位的指纹图谱分析,并通过柱色谱对其中的化学成分进行分离纯化,根据所得化合物的核磁、质谱数据及与对照品对比鉴定化合物的结构。结果 W-4和E-4为芪苈强心胶囊的强心、利尿及扩血管等活性部位,建立了其HPLC-PDA/ELSD的指纹图谱,并从中分离鉴定了11个化合物,分别为毛蕊异黄酮-7-O-β-D-葡萄糖苷(1)、人参皂苷Re(2)、人参皂苷Rg1(3)、杠柳苷G(4)、人参皂苷Rf(5)、人参皂苷Rb1(6)、人参皂苷Rc(7)、人参皂苷Rb2(8)、丹酚酸B(9)、山柰酚-3-O-芸香糖苷(10)和丹酚酸A(11)。结论明确了芪苈强心胶囊强心、利尿及扩血管的活性部位,11个化合物均为首次从芪苈强心胶囊中得到的化合物,这些化合物在活性部位中的指认为提供胶囊的质量标准奠定了基础,同时为阐述芪苈强心胶囊的药效物质打下了基础。 Objective To study the chemical components of active fractions in Qili Qiangxin Capsula. Methods The active fractions were obtained by means of solvent extraction, chromatography on absorbent resin, and activity screening. The fingerprints of active fractions were analyzed by HPLC-PDA/ELSD. The compounds were isolated by chromatography on silica gel and ODS repeatedly and their structures were elucidated on the basis of chemical methods and spectral analyses. Results The HPLC-PDA/ELSD fingerprints of active fractions (W-4 and E-4) with cardiac, diuretic, and vasodilator effects were esta-blished. The eleven compounds were obtained from them. They were identified as calycosin-7-O-β-D-glucopyranoside(1),ginsenosides Re(2), ginsenoside Rg1(3),periplocoside G(4), ginsenosides Rf(5), ginsenosides Rb1(6),ginsenosides Rc(7), ginsenosides Rb2 (8),salvianolic acid B(9),kaempferol-3-rutinoside(10),and salvianolic acid A (11). Conclusion The eleven compounds are identified from Qili QiangxinCapsula for the first time. This study establishes the foundation for improving the quality criteria and explaining the mechnism of Qili Qiangxin Capsula.
