摘要
目的 对改良化学氧化法和钒酸盐氧化法测定血清总胆红素和直接胆红素进行方法学评价.方法 采用改良化学氧化法测定血清总胆红素及直接胆红素值,与钒酸盐氧化法进行比较,并对改良化学氧化法的重复性试验、线性试验、干扰试验、回收试验进行评价.结果 改良化学氧化法和钒酸盐氧化法有很好的相关性 测定总胆红素的线性可达706.0 μmol/L,直接胆红素可达412.0 μmol/L 总胆红素、直接胆红素日内重复性变异均<3.0%,日间重复性变异均小于4.0% 总胆红素及直接胆红素的回收率平均分别为99.5%和99.75% 溶血、脂血、低浓度维生素C(<0.3 g/L)对测定无明显干扰.结论
Objective To compare improve chemical oxidation method and vanadate oxidation method in determination of total bilirubin and direct bilirubin in serum. Methods As comparing with the vanadate oxidation method, contents of total bilirubin and direct bilirubin in serum were assayed by the improve chemical oxidation method, and it was performed that the precision test, linear test, recover test and interfere test for the improve chemical oxidation method. Results There was a good correlation between the improve chemical oxidation and vanadate oxidation method. The linear range of total bilirubin was as high as 706. 0μmol/L,while that of direct bilirubin was as 412.0μmol/L. The daily variation of total bilirubin and direct bilirubin were less than 3.0%, and the day variation of total hilirubin and direct bilirubin were less than 4. 0%. The average recovery rate of total bilirubin and direct bilirubin was 99. 5% and 99. 75% , respectively. Hemoglohin,lipidemia and low - concentration vitamin Cless than 0. 3g/L did not significantly interfere with the detection. Conclusion The improve chemical oxidation method could be used in routine measuring for total biliruhin and direct hilirubin.
