富血小板血浆对人牙周膜成纤维细胞在脱矿病变牙根表面增殖及分化的影响 被引量:3

摘要 目的研究不同浓度的富血小板血浆(Platelet-Rich Plasma,PRP)对人牙周膜成纤维细胞(Humanperidontal fibroblasts,hPDLFs)在脱矿的病变牙根表面的附着及增殖情况,以探讨富血小板血浆在促牙周组织再生中的作用。方法选用组织块培养法培养人牙周膜成纤维细胞,选第五代细胞用于实验,制备病变的牙根片并进行脱矿处理。采用二次离心法分离全血,获取PRP,并配制为不同浓度的PRP待用。将制备的牙根片分组植入96孔板,分为包被PRP组和不包被组,对照组不加PRP,实验组加入不同浓度的PRP,用MTT法观察细胞附着及增殖的情况。结果实验组与对照组相比差异具有显著性,实验组10%~20%浓度组间相比差异具有显著性,20%~30%组间相比差异无显著性。结论适量PRP能促进人PDLFs在脱矿的病变牙根表面的附着及增殖,当PRP浓度为20%浓度达到最佳,包被PRP疗效更好。 Objective To investigate the effect of different concentrations of platelet-rich plasma(PRP) on cell proliferation and differentiation of human periodontal ligament fibroblasts((PDLFs) on the demineralized root surface of periodontitis affected teeth.Methods Human PDLFs were cultured from tissue explants and the 5th generation cells were used in this experiment.The cementum slices of periodontitis affected teeth were used and the surfaces were demineralizaed with EDTA.PRP was isolated by two-step centrifugation and was prepared in various concentrations.The cementum slices of periodontitis affected teeth were put on the 96-well tissue culture plates.The 5th generation cells were co-cultured with PRP at different concentrations:blank group,10% PRP group,15% PRP group,20% PRP group,and30% PRP group.Attachment and proliferation of cells were analyzed by MTT assay.Results Attachment and proliferation of PDLFs on cementum slices were significantly different between the experiment group and the control group in the experiment,there were significant difference between the 10% PRP group and 20% PRP group,there were no significant difference between the 20% PRP group and 30% PRP group.Conclusions PRP could promote the attachment and increase of PDLFs on the demineralized root surface of periodontitis affected teeth.The proliferation and differentiation of PDLFs in 20% PRP group were the highest.
