摘要
目的分析钙黏蛋白(Cadherin)8在新生野生型小鼠和Reeler小鼠大脑视觉皮层及海马齿状回中的表达变化,探讨Cadherin 8与Reelin蛋白之间可能的联系。方法实验动物共分2组,观察组为新生Reeler小鼠,对照组为新生野生型小鼠,Thionin染色和原位杂交法显示2组小鼠大脑视觉皮层与海马中的组织形态及Cadherin 8 mRNA的分布情况。结果对照组小鼠海马区由海马本部(CA1-CA3)和齿状回的双C型勾连结构组成,Cadherin 8 mRNA主要表达在大脑视觉皮层和海马中,在视觉皮层中表达呈明显分层现象;与对照组相比,观察组小鼠大脑视觉皮层细胞分层消失、海马本部和齿状回形态变化显著,Cadherin 8 mRNA在大脑视觉皮层中的表达分散且减弱,但在海马齿状回中的表达却明显增加。结论 Reeler小鼠大脑视觉皮层及海马齿状回中Cadherin 8的表达随相应组织结构的形态改变而变化,Cadherin8的表达和定位与Reelin的功能有一定关系。
Objective To analyze the expression change of Cadherin 8 in the cerebral visual cortex and hippocampus of postnatal wild type mouse and Reeler mouse,and to investigate the potential relation between Cadherin 8 and Reelin protein. Methods The Reeler mice were divided into observation group,and the wild type mice were divided into control group. The morphology of the cerebral visual cortex and hippocampus in two groups as well as the distribution of Cadherin 8 mRNA were showed by Thionin staining and in situ hybridization method. Results The hippocampus of mice in control group was composed by double C link structure of CA1-CA3 and dentate gyrus. The main expressions of Cadherin 8 mRNA were lined in cerebral visual cortex and hippocampus in control group,and the expression in cerebral visual cortex showed clear stratification. Compared with the control group,the layers of the cerebral visual cortex were disappeared,and the morphology of hippocampus was clearly changed in observation group. The expression of Cadherin 8 mRNA in cerebral visual cortex became scattered and weaker,but the expression became clearly stronger in the dentate gyrus of hippocampus. Conclusion The expression changes of Cadherin 8 in the cerebral visual cortex and hippocampus of Reeler mouse are related with the changes of morphology of organization structure, and the expression and location of Cadherin 8 is related with the function of Reelin protein.
