摘要
目的观察转化生长因子(TGF)-β1不同作用时间对绒癌JEG-3细胞增殖及基质金属蛋白酶抑制剂(TIMP)-1的影响。方法以100μg/LTGF-β1处理JEG-3细胞,分别于不同时间收获细胞,采用MTT法于0、12、24、48h检测细胞的增殖活力,采用Western Blotting及RT-PCR技术于0、12、24、48、72h分析TIMP-1在蛋白和转录水平的表达情况。结果随着100μg/LTGF-β1作用时间的延长,各实验组的细胞增殖率逐渐升高,差异有统计学意义(P<0.05);TIMP-1蛋白和TIMP-1 mRNA的表达随着100μg/LTGF-β1作用时间的延长均呈明显的增加趋势(P<0.05)。结论绒癌JEG-3细胞对外源性的TGF-β1有良好的反应性并呈时间依赖性,100μg/LTGF-β1可促进JEG-3细胞增殖及TIMP-1蛋白及其mRNA的表达。
Objective To observe the effect of β1 transforming growth factor β1 (TGF β1) at different action times on cell proliferation and tissue inhibitor of metallopmteinase-1 (TIMP-1) expression in JEG-3 choriocarcinoma cell.Methods JEG-3 choriocarcinoma cells were treated with 100 μg/L TGF-β1,then the cells were respectively collected as different times.Cell proliferation was determined by using MTT(0, 12, 24 and 48 hours),and expression of TIMP-1 by Western Blotting and RT-PCR(0, 12, 24,48 and 72 hours).Results With the extension of TGF β1 action time,the ratio of cell proliferation in each group increased (P0.05 ),and TIMP-1 protein and TIMP-1mRNA were both upregulated significantly (P0.05).Conclusion JEG-3 choriocarcinoma cell can response to exogenous TGF β1 and there is a time-effect relationship.TGF β1 in 100μg/L can promote JEG-3 cell proliferation and expression of TIMP-1 protein and its mRNA.
