摘要
比较了3种RNA提取方法提取石榴果皮RNA的效果。结果表明:改良CTAB法提取的总RNAOD值在1.91左右,得率为48μg/g;Trizol法提取总RNAOD值在1.15左右,得率为86μg/g;试剂盒提取得到的总RNAOD值在1.24左右,得率为124μg/g。分别反转录为cDNA,根据不同植物花青素合成关键酶基因保守序列,设计简并引物进行RT-PCR,其中只有改良CTAB法得到的cDNA有扩增条带。表明改良CTAB法比较适合富含多糖和酚类物质石榴果皮总RNA的提取。
Three RNA extraction methods from pomegranate peel were compared.The result showed that the OD value of total RNA was 1.91 by modified CTAB,the yield was 48μg/g;the OD value of total RNA was 1.15 by Trizol method,the yield was 86μg/g;the OD value of total RNA was 1.24 by RNA extracted kit,the yield was 124μg/g;the cDNAs were got by reverse transcription.A pair of degenerate primers was designed based on the conserved domain of CHS that was one of the key genes of anthocyanidin synthesis of eight plants.Only the cDNA from the improved CTAB had one amplified band by RT-PCR,the others had no bands.So the improved CTAB method was suitable for RNA extraction of pomegranate peel.
