摘要
通过对hG CSF重组基因表达效率和工程菌遗传稳定性的比较 ,建立的一种全新的双质粒双重诱导的原核高效表达体系。构建了pGP1 2 pJGW1 hGCSF DH5α工程菌 ,经 42℃热诱导 ,hG CSF的表达率达 2 7%以上 ,经典质粒遗传稳定性实验 (140代 )证实 ,具有良好的遗传稳定性。
A novel double induced expression system containing plasmids pJGW1 hGCSF and pGP1 2 was constructed on the basis of study on the expression efficiency of recombinant hG CSF gene and the genetic stability of enginerring E.coli.After the recombinant bacterial strain,pGP1 2 pJGW1 hGCSF/DH5α,was induced at 42℃,27% of hG CSF was expressed.The recombinant bacterial strain had been undergone 140 passages,the result showed the good genetic stability of it.
