四妙君逸软膏用于肛肠病术后换药镇痛的药理学实验研究

Pharmacological study of Simiao Junyi Ointment in postoperative analgesic effect on anorectal diseases

目的:研究四妙君逸软膏的镇痛作用及其镇痛作用与阿片受体、β-内源性阿片肽(β-EP)、5-羟色胺(5-HT)的关系。方法:采用小鼠热板法、扭体法观察四妙君逸软膏对小鼠镇痛作用的影响;采用纳洛酮拮抗实验,醋酸致痛后检测小鼠血清β-EP和脑组织5-HT的含量,探讨四妙君逸软膏的镇痛作用机制。结果:四妙君逸软膏能够明显延长痛阈和抑制小鼠扭体反应,镇痛作用优于马应龙麝香痔疮膏。其镇痛作用能被纳洛酮部分拮抗,并能明显提高小鼠血清β-EP和脑组织5-HT的含量。结论:四妙君逸软膏能够提高小鼠血清β-EP和脑组织5-HT的含量,具有明确的镇痛作用。其镇痛作用与阿片受体、β-EP、5-HT有关。

Objective： To evaluate analgesic effect of Simiao Junyi Ointment,and to explore the relationship of its analgesic effect and opioid receptor,β-endorphin,5-hydroxytryptamine.Methods：Mouse hot-plate test and writhing experiments were used to observe the analgesic effect of Simiao Junyi Ointment on mice.Naloxone reversal test was used to detection the levels of serum β-EP and 5-HT in brain tissue after the acetic acid-induced pain,to explore the analgesic mechanism of Simiao Junyi Ointment.Results：Simiao Junyi Ointment can obviously prolong the pain threshold in mice and inhibite the mouse writhing response.It was better than Musk Hemorrhoids Ointment and Tetracaine Hydrochloride Jelly.The analgesic effect of Simiao Junyi Ointment can be partly antagonized by naloxone.Simiao Junyi Ointment could obviously improve the content of β-endorphin in serum and brain 5 - hydroxytryptamine.Conclusion：Simiao Junyi Ointment can regulate up the content of β-endorphin in serum and brain 5 – hydroxytryptamine,its analgesic effect may be related to opioid receptor,β-endorphin and 5-hydroxytryptamine.
