摘要
目的为杏丁注射液治疗肾脏应激性损伤提供理论依据。方法将对数生长期肾小管上皮NRK52E细胞随机分为八组。对照组不干预;H2O2组加入H2O2使终浓度为400μmol/L,杏丁1、2、3组分别加入杏丁注射液使终浓度分别为0.4、4、40μg/ml,H2O2+杏丁1、2、3组分别加入H2O2(终浓度同前)及杏丁注射液(终浓度同前)。流式细胞仪检测各组细胞凋亡率,RT-PCR方法检测SurvivinmRNA表达。结果 H2O2组细胞凋亡率明显高于对照组及杏丁1、2、3组;H2O2+杏丁2、3组凋亡率明显低于H2O2组;H2O2+杏丁组SurvivinmRNA表达明显高于H2O2组。结论杏丁能显著抑制H2O2诱导的肾小管上皮细胞凋亡,其作用机制可能与其可增加Survivin表达有关;杏丁注射液可用于肾脏应激性损伤的治疗。
Objective To provide the theroy base of Xingding injection on treating stress-induced renal injure. Methods The renal tubular epithelial cells in logarithmic phase were randomly divided into eight groups. The control group was not interfered; The H2O2 group was treated with 400 μmol/L H2O2,the Xingding 1,2,3 groups were added Xingding injection with the concentration of 0. 4,4 and 40 μg/ml,the H2O2 + Xingding 1,2,3 groups were added with H2O2 and Xingding injection. Apoptosis was assessed by flow cytometry,the expression of Survivin mRNA was detected by RT-PCR. Results The apoptosis rate of H2O2 group was significantly higher than that of control group and Xingding 1,2,3 groups. The apoptosis rate of H2O2 + Xingding group was significantly higher than that of H2O2 group; the expression of Survivin mRNA of H2O2 + Xingding group was significantly higher than that of H2O2 group. Conclusions Xingding can inbibit the apoptosis of renal tubular epithelial cell induced by H2O2,which maybe associated with the upregulated expression of Survivin; Xingding injection can be used to treat the stress-induced renal injure.
