摘要
目的研究TGFα,EGFR反义寡聚核苷酸抑制大肠癌HR8348细胞株恶性增殖的作用.方法采用23个碱基组成的TGFα反义寡聚核苷酸和21个碱基组成的EGFR反义寡聚核苷酸以及21个碱基组成的对照寡聚核苷酸作用于人大肠癌HR8348细胞,通过细胞生长抑制实验,3HTdR掺入,mRNA斑点杂交及细胞周期分析以确定其对大肠癌细胞的增殖抑制作用和作用环节.结果TGFα,EGFR反义寡聚核苷酸与对照寡聚核苷酸相比较,均能有效抑制HR8348细胞的增殖(P<001),DNA合成(P<001)和mRNA(P<001)的表达,并能有效地延缓HR8348细胞由G0/G1期向S期的转化。
AIM To study the inhibitory effect of transforming growth factor alpha(TGF α), epidemal growth factor receptor(EGFR) anti sense oligodeoxynucleotides on the malignant proliferation of the HR8348 cell line . METHODS TGFα, EGFR anti sense oligodeoxynucleotides, composed of 23 and 21 bases, respectively, and indifferent oligodeoxynucleotides were used to affect the HR8348 cells. The cell growth inhibition, 3H TdR incorporation,mRNA hybridization and the cell cycle analyses were made to identify the inhibitory effect and mechanism of TGFα, EGFR anti sense oligodeoxynucleotides. RESULTS TGFα, EGFR anti sense oligodeoxynucleotides both could inhibit the proliferation of HR8348 cell; the DNA synthesis; the mRNA expression; and delay the transition period of G 0/G 1 phase to S phase. CONCLUSION TGFα, EGFR anti sense oligodeoxynucleotides both could inhibit the malignant proliferation of the HR8348 cell effectively.
