热带假丝酵母两态型的控制研究 被引量:6

摘要 通过对热带假丝酵母在不同培养基上的培养,研究了培养温度、pH值、脯氨酸浓度及培养时间对酵母两种形态的影响。通过正交试验得出:最有利于菌丝型生长的条件是:YX培养基、42℃、pH值6.5、接种量5%、木糖含量2%、24h补加10mmol/L脯氨酸、215r/min摇床培养,验证试验表明其最大丝化率为73.39%,平均丝化率为67.29%;最有利于酵母型生长的条件是:YM培养基、pH值4.5、27℃、215r/min摇床培养,其最大丝化率仅为1.14%,平均丝化率为0.08%。 Effects of medium, incubation temperature, pH value, concentration ofproline on dimorphism of Candida tropicalis were studied. The optimum conditions for mycelial form were obtained by orthogonal experiments, which were YX medium with addition of 2% xylose and 10mmol/L proline, pH 6.5, incubation under 42℃ for 24h with inoculum of 5% with shaking at 215r/min. Under the optimum conditions, the maximum and average mycelial rate was 73.39% and 67.29%, respectively. The optimum conditions for yeast-like growth were YM medium, pH value 4.5, incubation under 27℃, with shaking at 215r/rain. And under the conditions, the maximum and average mycelial rate was 1.14% and 0.08%, respectively.
