以长江流域棉花新品系川239为材料,系统比较了四种激素组合(2,4- D+ KT,NAA+ KT,IAA+ KT, ZT)对愈伤组织诱导和体胚发生的影响,对获得大量再生植株的条件进行了研究,获得了“川239”再生植株。
The Chuan 239 planted in the Yangtze River Cotton Region is a recalcitrant cotton variety for somatic embryogenesis. To achieve somatic embryogenesis from it, several kinds of hormone and subculture methods were investigated. The medium MSB containing 2,4 D 0 05mg·L -1 +KT0.1mg·L -1 or NAA 4mg·L -1 +KT1mg·L -1 was good for callus initiation but the callus didn't produce embryos after subculture. The medium MSB Containing IAA0 5mg·L -1 +KT0 1mg·L -1 or ZT0 1mg·L -1 only yielded a very low inducing frequency and growth speed of callus, but all the callus were embryogenic. When the embryogenic callus were subcultured with SM 1(MSB+IAA0 1mg·L -1 +KT0 1mg·L -1 ) and SM 2(MSB+KT0 1mg·L -1 +KNO 3 1 9g·L -1 ) alternatively, the growth of nonembryogonic callus were retarded and somatic embryos were produced. Suspension culture promoted somatic embryogenesis. For germination of somatic embryos, the medium SH with 0 5g·L -1 active coal was better than that of medium MSB with BA 0 1mg·L -1 and IAA0 1mg·L -1 or hormone free MSB medium. Regenerated plants from somatic embryos were obtained. The strategy to achieve somatic emdbryogenesis from recalcitrant cotton varieties was discussed.
