大鼠酒精性肝病形成中瘦素基因的表达及三七对其的影响 被引量:2

The effect of Sanchi on the expression of hepatic LEPTIN OB-RB and TGF-β1 mRNA in alcoholic liver disease rats

摘要 目的:观察酒精性肝病形成中大鼠肝组织瘦素及其受体、TGF-β1基因的mRNA表达的动态变化,三七干预的影响,探讨瘦素在酒精性肝病形成中的作用及其三七抗酒精性肝病的作用机制。方法:以白酒-玉米油-吡唑混合液灌胃建立酒精性肝病动物模型,同时以高、低剂量的三七及硫普罗宁进行干预,分别在4、8、14周末光镜观察大鼠肝脏组织学变化情况,RT-PCR检测肝组织瘦素及其受体、TGF-β1基因的mRNA表达的动态变化。结果:随着脂肪变及炎症程度的加重,肝组织瘦素及其受体、TGF-β1基因的mRNA逐渐增强,肝组织中瘦素及其受体mRNA表达水平与肝脏脂肪变、炎症程度以及TGF-β1 mRNA表达均呈明显正相关;应用三七及硫普罗宁干预14周后,大鼠肝组织脂肪变及炎症程度明显减轻,同时肝组织瘦素及其受体、TGF-β1基因的mRNA表达的明显下降。结论:瘦素及其受体参与大鼠酒精性肝病的发生和发展,三七通过抑制肝组织局部Lep-tin、OB-R以及TGF-β1的基因表达水平促进肝组织病理的改善,阻止酒精性肝病的慢性化进程,这可能是其防治酒精性肝病的重要机制之一。 Objective : To observe the effect of Sanchi on changes of expression of hepatic leptin , OB - RB and TGF-β1 mRNA in rat with alcoholic liver disease, and to explore the mechanism of Sanchi in alcoholic liver disease. Methods: The model of Alcoholic liver disease in rats was induced with mixture of alcohol, eom oil, and pyrazole inl4 weeks. Rats in the high-dose Sanchi, the low-dose Sanehi, and the Tiopronin group were treated with different medieine. To observe the degree of liver steatosis and inflammation at the end of4th 8th 14th week; leptin,OB - PuB and TGF-β1 mR- NA were dynamically examined by RT- PCR method. Results: With the prolonging of drinking time, the expression of heparle leprln,OB -RB and TGF TGF-β1 mRNA were gradually increased , at the same time ,the degrees of hepatic steatosis and inflammation were further severe. The expression of leptin, OB - RB and TGF-β1 mRNA were lowed obviously in Sanqi and Tiopronin group at the end of 14th week and meanwhile Sanqi and Tiopronin can improve the degree of liver steatosis and inflammation. Conclusion: Sanehi can markedly reduce the expression of leptin, OB - RB and TGF-β1 mRNA in rat with alcoholic liver disease. This may serve as one of the critical mechanisms through which Sanchi efficiently prevents from alcoholic liver disease.
