高效液相色谱法测定川产土牛膝中齐墩果酸的含量

HPLC determination of oleanolic acid in achyranthes produced in Sichuan

目的:建立高效液相色谱法(HPLC)测定川产土牛膝中齐墩果酸含量的方法。方法:采用HPLC法,色谱柱为Dikma Kro-masil C18(250 mm×4.6 mm,5μm);流动相为甲醇-水(92∶8);检测波长:210 nm;柱温:30℃;流速:1.0 mL.min-1;进样量:10μL。结果:齐墩果酸在1～10μg范围内具有良好的线性关系(r=0.9999),平均回收率为96.97%,RSD为1.02%。结论:该方法简便、快速、准确、具有良好的重复性和回收率,可作为齐墩果酸的定量分析方法。

Objective：To establish a HPLC method for the determination of oleanolic acid in achyranthes produced in Sichuan.Methods：The separation was carried out on the Dikma Kromasil C18 column（250 mm×4.6 mm,5 μm）.The mobile phase was methanol-water（92：8）.The detective wavelength was set at 210nm.The column temperature was 30℃ and the flow rate was 1.0 mL·min-1 with the sample volume of 10μL.Results：The calibration curve showed good linearity over the range of 1-10 μg（r=0.9999）.The average recovery rate was 96.97% with RSD 1.02%.Conclusion：This method is simple,rapid,accurate with good repeatability and good recovery,which can be used as the quantitative analysis method of oleanolic acid for achyranthes produced in Sichuan.
