牛膝多糖对糖尿病大鼠视网膜细胞凋亡的影响 被引量:11

摘要 目的探讨牛膝多糖(achyranthes bidentata blume polysaccharides,ABPS)对糖尿病大鼠视网膜病变的保护作用及其作用机制。方法将Wistar大鼠随机分为5组,正常对照组、糖尿病模型组、ABPS高、中、低剂量组(300mg.kg-1.d-1、200mg.kg-1.d-1、100mg.kg-1.d-1)。8周后,琼脂糖凝胶电泳检测细胞凋亡,RT-PCR观察bax、survivin基因的表达。结果与糖尿病模型组比较,ABPS各组视网膜细胞凋亡明显减少,survivin表达上调,bax表达降低。结论 ABPS对糖尿病大鼠视网膜组织保护作用的机制可能与抑制视网膜组织细胞凋亡及上调survivin,降低bax基因表达有关。 OBJECTIVE To investigate the effect and mechanism of achyranthes bidentata blume polysaccharides(ABPS)on retinal tissue of diabetic rats.METHODS Wistar rats were randomly divided into normal control group,diabetic model group and diabetic model treated with 300,200 and 100mg·kg-1·d-1 ABPS groups.After 8 weeks,the number of apoptotic cells was checked by electrophoresis and the gene expressions of bax and survivin were determined by RT-PCR.RESULTS In comparison with diabetic model group,the number of apoptotic cells in retinal tissue of ABPS groups was decreased.And ABPS could effectively decrease the level of bax gene expression and improve the level of survivin gene expression.CONCLUSIONS ABPS had protective effect on retinal tissue of diabetic rats.Inhibiting the apoptosis cells,regulating bax and survivin gene expressions may be the possible mechanism of this protective defect.
