家兔体内用硅胶囊构建自体复合皮肤的研究 被引量:1

摘要 目的:观察在体内构建类似于皮肤结构的复合皮的可行性。方法:大耳白兔10只,于其背部左右侧皮下各埋植一球形的医用硅胶扩张囊,手术1周后,在扩张囊周围形成纤维包囊,此时将原代培养的皮肤上皮细胞悬液经皮下埋置的管道注入扩张囊和纤维包囊腔隙之间。设对照组。2周后取出扩张囊,肉眼及光镜观察纤维包囊表面是否有上皮形成。结果:肉眼观实验组包囊透明度稍低于对照组之外,未见明显区别;光镜下对照组未见有上皮组织覆盖,而实验组可见类似于假复层鳞状上皮细胞层。结论:用上述方法体内构建类似于皮肤结构的复合皮有可行性,有望将纤维包囊连同其表面生长的上皮细胞层用于创面覆盖。 Objective:To observe the feasibility of constructing the composite skin in vivo whose structure is similar to the skin.Methods:A medical silicone expander was implanted subcutaneously in the back,on both sides symmetrically in each of 10 rabbits.1 w after surgery,fibrous cyst formed around the expander and cultured isogeneic rabbit keratinocytes suspension was injected subcutaneously by the pipeline into the room between fibrous cyst and expander.A control group was set up and the expander was removed after 2 w to see whether the epithelialization could be found on the surface of fibrous cyst by macroscopic and microscopic observation.Results:Transparency of cyst in the experimental group was slightly lower than that of the control group by macroscopic observation,and no other significant difference was found.There was no epithelialization on the surface of fibrous cyst in the control group by microscopic observation whereas structure similar to the epithelializatified on the surface of fibrous cyst in the experiment group could be observed.Conclusion:The composite skin in vivo whose structure is similar to the skin can be constructed with the above,epithelium with the fibrous cyst is feasible for wound coverage.
