TTC染色识别家猪急性心肌梗死模型中存活心肌的研究 被引量:3

TTC dyeing detecting viable myocardium in pig models with acute myocardial infarction

摘要 目的:应用经皮腔内冠状动脉成形术球囊堵闭猪冠状动脉建立急性心肌梗死动物模型,并通过TTC染色识别急性心肌梗死动物模型中存活心肌和梗死心肌。方法:普通家猪13只,麻醉气管插管后行冠状动脉造影术,置入经皮腔内冠状动脉成形术球囊至冠状动脉左前降支远端,堵闭血流60~90 min,建立家猪心肌梗死模型。处死试验动物取出心脏,将心肌组织切成薄片后采用TTC染色法识别存活心肌。结果:家猪均完成冠状动脉左前降支远端的封堵,2只家猪分别在堵闭45 min,65 min时因心室颤动死亡,存活的11只家猪均成功建立急性心肌梗死模型,TTC染色显示正常心肌88个节段,梗死心肌49个节段,存活心肌39个节段。结论:运用经皮腔内冠状动脉成形术球囊封堵冠状动脉可成功建立猪急性心肌梗死模型,通过TTC染色能识别出心肌梗死模型中的梗死心肌与存活心肌。 Objective To identify viable myocardium and infarct myocardium with TTC dyeing technique by establishing pig models with acute myocardial infarction by using percutaneous transluminal coronary angioplasty sacculus.Methods Thirteen pigs received coronary angiography by inserting percutaneous transluminal coronary angioplasty sacculus to the end of the left anterior descending coronary artery after anesthesia tracheal intubation.Then the blood flew was blocked for 60 to 90 minutes,and the pig models with acute myocardial infarction were established.The pigs were sacrificed and the hearts were taken out,then the heart tissue was cut to slices and recognized the viable myocardium with TTC dyeing.Results All pigs were completed the blocking of distal end of the left anterior descending coronary artery,in which two pigs died because of ventricular fibrillation in 45 and 65 minutes after blocking.The other 11 pigs were established the model of acute myocardial infarction successfully.TTC dyeing indicated that normal myocardium was in 88 segments,infarct myocardium was in 49 segments and viable myocardium was in 39 segments.Conclusion To explore coronary artery by using PTCA sacculus can establish the pig models of acute myocardial infarction successfully,and the TTC dyeing detecting can finally identify viable myocardium and infarct myocardium.
