反相高效液相色谱法测定地塞米松软膏中地塞米松含量

Content Determination of Dexamethasone in Dexamethasone Ointment by RP-HPLC

目的 建立反相高效液相色谱（RP-HPLC）法测定地塞米松软膏中地塞米松的含量.方法采用Sinochrom ODS-BP（4.6 mm×250 mm,5 μm）色谱柱,甲醇-水（80：20）为流动相,检测波长240 nm,流速1.0 mL·min-1,进样量20 μL.结果地塞米松在0.25～2.50 μg范围内呈良好线性关系（r＝0.999 9）,平均回收率100.1%,RSD＝0.75%（n＝9）.结论 该方法简便、快速、分离度好、结果稳定,可用于地塞米松软膏的质量控制.

Objective To develop a method for content determination of dexamethasone in dexamethasone ointment by RP-HPLC. Methods The separation was performed on Sinochrom ODS-BP（4.6 mm×250 mm,5 μm）colunm with methanol： water（80 ： 20） as a mobile phase,and with a wavelength of UV detector at 240 nm. The flowing rate was 1.0 mL·min-1 and the loading volumn was 20μL. Results The linear range of dexamethasone was within 0.25-2.50 μg （r = 0. 999 9）. The average recovery of dexamethasone was 100. 1% with RSD as 0.75% （n = 9）. Conclusion The method is simple, rapid, accurate and reliable,which can be applied to the quality control of dexametbasone ointment.
