配伍前后黄连解毒汤中4种有效成分含量的对比研究 被引量:10

Comparative Study of 4 Active Constituents in Huanglian Jiedu Decoction before and after Mixing

摘要 目的:建立同时测定黄连解毒汤中栀子苷、盐酸小檗碱、盐酸巴马汀和黄芩苷4种有效成分含量的方法,探讨配伍对4种成分含量的影响。方法:采用反相高效液相色谱法。色谱柱为Diamonsil C1(8250mm×4.6mm,5μm),流动相为乙腈-0.1%冰乙酸(线性梯度洗脱),流速为1.0mL.min-1,柱温为40℃,检测波长为260nm。结果:栀子苷、盐酸小檗碱、盐酸巴马汀和黄芩苷进样量分别在0.1250~0.6250、0.0500~0.2500、0.0375~0.1875、0.0450~0.2250μg范围内与各自峰面积积分值呈良好的线性关系(r分别为0.9993、0.9992、0.9994、0.9994);平均加样回收率分别为97.54%、96.07%、104.00%、96.79%,RSD分别为0.78%、0.78%、0.74%、0.83%(n=6)。黄连解毒汤合煎液中4种成分的含量均低于相同处方量的单煎液。结论:方剂配伍后化学成分的含量并不是组方药味的简单叠加,而有其内在的配伍规律。本研究结果可为黄连解毒汤配伍规律的研究提供参考。 OBJECTIVE:To establish a method for simultaneous determination of jasminoidin,berberine hydrochloride,palmatine hydrochloride and baicalin in Huanglian jiedu decoction(HLJDD),and to explore the influence of compatibility on their contents.METHODS:RP-HPLC method was adopted.The samples were separated on a Diamonsil-C18(250 mm×4.6 mm,5 μm) column with acetonitrile-0.1% glacial acetic acid as mobile phases(gradient elution) at flow rate of 1.0 mL·min-1.The column temperature was at 40 ℃ and the detection wavelength was 260 nm.RESULTS:The linear ranges of jasminoidin,berberine hydrochloride,palmatine hydrochloride and baicalin were 0.125 0~0.625 0 μg,0.050 0~0.250 0 μg,0.037 5~0.187 5 μg,0.045 0~0.225 0 μg,respectively(r=0.999 3,0.999 2,0.999 4 or 0.999 4).Average recoveries were 97.54%,96.07%,104.00% and 96.79%.RSDs were 0.78%,0.78%,0.74%,0.83%(n=6).The contents of 4 active components in HLJDD were all lower than those of same formula dose of single component decoction.CONCLUSION:The contents of chemical compositions in HLJDD are not a simple addition of their contents in single medicine,but have their own internal compatibility law.This article provides a reference for the study of compatibility regularity of HLJDD.
