摘要
将结核杆菌ESAT-6 和MPT-64 的编码基因分别插入真核表达载体JW 4303 构建重组质粒,用重组质粒对4 周龄的BALB/C和F1 代小鼠进行肌肉和皮内免疫,于末次免疫后10 天采血并分离血清,进行ELISA 检测。结果表明,小鼠能产生较强的体液免疫,特异性IgG 平均水平分别为参考血清的2.20 倍和3.19 倍,且对两种品系小鼠进行的相同途径的免疫,其抗体水平差异不显著;而不同途径的重组质粒免疫,抗体水平略有差异。
ESAT 6 and MPT 64 genes of TB were inserted into eukaryotic expression vector JW4303 respectively.Two recombinant plasmids were immunized with BALB/C and F1 mice intramuscularly and subcutaneously and produced positive humoral immune effect.The antibody response injected by intramuscular was higher slightly than by subcutaneous.There was no significant difference of antibody levers in BALB/C and F1 mice after immunication of recombinant plasmids.
