摘要
为了对产碱性蛋白酶的地衣芽孢杆菌D.1的培养条件进行优化,利用10L发酵罐,采用正交设计L9(3^4)试验,对培养温度、pH值、搅拌转速、通气量4条件进行优化,得到地衣芽孢杆菌D-1发酵产碱性蛋白酶的最优培养条件为:培养温度37.0℃,pH值7.5,通气量4L/min,搅拌转速300r/min。利用最优条件组合进行验证实验表明,该培养条件下地衣芽孢杆菌D-1碱性蛋白酶的酶活为91.26U/mL。
In order to optimize culture condition for alkaline protease production by Bacillus licheniformis D-1, the culture temperature, pH, stirring speed and air flow rate during fermentation were optimized by the L9 (34) orthogonal test in 10 L bioreactor. The optimized culture conditions were determined as a culture temperature of 37 ℃ , a pH of 7, 5, an air flow rate 4 L/min, and a stirring speed of 300 r/min. Under these optimization conditions, the confirmative experiment shows that the activity of alkaline protease produced by Bacillus licheniformis D-1 reached 91.26 U/mL.
