摘要
将7日龄的黑白花奶牛胚胎,采用简易冷冻仪,以液氮作冷冻源的快速冷冻、解冻法对胚胎进行冷冻.甘油作冷冻保护剂,以1℃/min的速度由室温降至-7℃,在-7℃下绣发结晶,并在-7℃下维持5分钟;又以0.3℃/min降到-10℃,最后以0.5℃/min速度降至-30℃,平衡10分钟后,胚胎直接投入液氮罐申保存。需移植时,直接在37℃温水中解冻。本试验解冻了25枚胚胎,取1,2级胚胎15枚,除去甘油后,将15枚胚胎用凯氏枪输卵器分别移植给15头同期受体的的母牛,现已产下3头牛犊.结果表明,采用简易冷冻仪、以液氮作冷冻源的快速冷冻、解冻的胚胎可以成活,且有利在生产实践中推广应用。
Seven-day Black and White cow embryos were speed-frozen with a low-cost freezer by using nitrogen as a refrigerant and glycerol as a protectant. The cooling process was started at ambient temperature. First the temperature was dropped to -7℃ at 1℃/ min and then crystalization was induced; the temperature was held at -7℃ for 5 minutes; again the temperature was dropped from -7℃ to -10℃ at 0.3℃/ min and finally from -10℃ to -30℃ at 0.5℃/min. After 10 minutes remaintance, the containers of embryos were immerged directly into liquid nitrogen. They were thawed by putting them directly into warm water at 37℃. In this experiment 25 embryos were thawed, of which 15 thawed embryos in grade 1 and grade 2 were collected. After getting rid of glycerol 15 thawed embryos were transfered to 15 simultaneous recipient cows with a catheter. 15 of which were checked and 3 recipients were pregnant and calves have been born. It showed that embryos which are speed-frozen and thawed in liquid nitrogen with a low-cost freezer can be survived. It is beneficial to popularization and application of ET in production.
