摘要
近年来 ,随着一些先进的细胞和分子生物学技术的引进与建立 ,我国毒理学研究整体水平得以大大的提高。现介绍几种我国已开展的细胞和分子生物学新技术的原理及其在毒理学领域中的应用 ,包括 :差异显示技术、单细胞凝胶电泳技术、穿梭质粒 p Z189和 p Sp189转染技术、转基因动物和转基因细胞、荧光原位杂交和流式细胞技术。这些技术具有终点明确 ,敏感性高等特点 ,在检测各种不同类型的 DNA损伤和基因突变 。
Following introduction and establishment of some advanced technologies on cellular and molecular biology,the overall level of toxicological research in China has been greatly improved in recent years.Both principle and toxicological application of new technologies on cellular and molecular biology,including differential display,single cell gel electrophoresis(comet assay),shuttle plasmid(pZ189 and pSp189) transfection,transgene animals and cells,fluorescence in situ hybridization (FISH)and flow cytometry,developed and carried out in China were reviewed.These technologies are characterized with clear endpoints and higher sensitivities,They are very important values in both detecting various types of DNA damages and gene mutations and exploring the molecular mechanisms of cellular toxicity and carcinogenesis.\;
