摘要
研究苯甲酸、肉桂酸和曲酸复合使用对酪氨酸酶活性的抑制以及抑制剂间的相互作用。采用酪氨酸酶L-多巴速率氧化法,观测不同比例的复合抑制剂对酪氨酸酶二酚酶的活性影响,以研究药物相互作用的等效线分析法和相互作用指数评价复合抑制剂的相互作用效应。结果显示在等效于酶活性抑制50%的条件下,苯甲酸与曲酸质量比为7.5∶1,相互作用指数为0.42;肉桂酸与曲酸质量比为3.8∶1,相互作用指数为0.56;苯甲酸、肉桂酸和曲酸质量比为7.5∶3.8∶1,相互作用指数为0.18。说明苯甲酸、肉桂酸和曲酸复合使用对酪氨酸酶二酚酶活性抑制的相互作用主要表现为协同效应,协同程度与复合抑制剂质量浓度和比例有关。
In order to study the inhibitive effects of combination of Benzoic Acid,Cinnamic Acid and Kojic Acid on tyrosinase of mushroom,the effects of combination inhibitor on diphenolase activity were observed by measuring the oxidation rate of L-dopa.Isobolographic plot and interaction index were employed to evaluate the inhibitors synergistic effects.The results show that in 50% inhibitory rate of enzyme activity the interaction index was 0.42 when combined with Benzoic Acid and Kojic Acid at the ratio 7.5∶1,the index was 0.56 when combined with Cinnamic Acid and Kojic Acid at the ratio 3.8∶1,and the interaction index was 0.18 when combined with Benzoic Acid,Cinnamic Acid and Kojic Acid at the ratio 7.5∶3.8∶1.Inhibitive effects of the combinaion of Benzoic Acid,Cinnamic Acid and Kojic Acid mainly produce synergism,the mixture concentration and mixing ratio have a great influence on the synergistic effects.
