摘要
目的探讨经肝动脉化疗栓塞术(TACE)联合姜黄素治疗兔VX-2肝癌对血管内皮生长因子VEGF表达的影响。方法建立兔VX-2肝癌模型40只,按随机数字表法分成生理盐水组(对照组)、姜黄素灌注组(HA组)、碘化油栓塞组(TAE组)和姜黄素碘化油乳剂栓塞组(TACE组),每组10只。对照组:经肝动脉缓慢注入1 mL生理盐水;HA组:经肝动脉缓慢注入姜黄素5 mg.kg-1;TAE组:经肝动脉缓慢栓塞碘化油1 mL;TACE组:经肝动脉缓慢栓塞姜黄素碘化油乳剂1 mL。治疗后2周处死实验兔,取出肿瘤标本。采用免疫组织化学法和Western印迹杂交法测定新生灶中VEGF蛋白的表达。结果免疫组织化学法VEGF蛋白的表达水平:对照组为0.582±0.181,HA组为0.524±0.277,TAE组为0.874±0.199,TACE组为0.581±0.190;Western印迹杂交法VEGF蛋白的表达水平:对照组为46.49±12.78,HA组为47.54±14.51,TAE组为83.40±18.02,TACE组为54.21±15.70。TAE组中VEGF蛋白的表达明显高于对照组、HA组和TACE组(P<0.05),对照组、HA组、TACE组之间VEGF蛋白的表达差异无统计学意义(P>0.05)(2种测定方法结果相一致)。结论姜黄素联合碘化油栓塞兔VX-2肝癌模型后能有效地降低VEGF蛋白的表达,而单纯姜黄素经肝动脉灌注不能有效地降低VEGF蛋白的表达。
Objective To investigate the effects of transcatheter arterial chemoembolization(TACE) combined with curcumin on the expression of vascular endothelial growth factor(VEGF) in rabbit VX-2 liver tumors.Methods Forty VX-2 tumor-bearing rabbits were randomly divided into four groups,with 10 rabbits in each group.Control group,HA group,TAE group and TACE group received hepatic arterial infusion of normal saline(1 mL),hepatic arterial injection of curcumin(5 mg·kg-1),hepatic artery embolization with lipiodol(1 mL),and hepatic artery embolization with lipiodol mixed with curcumin(1 mL),respectively.The rabbits were sacrificed and the tumors were taken 2 weeks after embolism.Immunohistochemistry and Western blotting were used to detect the expression of VEGF.Results The levels of VEGF expression measured by immunohistochemistry were 0.582±0.181,0.524±0.277,0.874±0.199 and 0.581±0.190 in control,HA,TAE and TACE group,respectively.The levels of VEGF expression detected by Western blotting were 46.49±12.78,47.54±14.51,83.40±18.02 and 54.21±15.70 in control,HA,TAE and TACE group,respectively.The expression of VEGF in TAE group was significantly higher than that in the other three groups(P0.05),and there were no significant differences among them(P0.05).Conclusion TACE combined with curcumin can obviously depress the expression of VEGF in rabbit VX-2 liver tumors.
