摘要
目的:观察葛根素对糖尿病大鼠肾脏PDGF-BB表达的影响。方法:将SD大鼠随机分为正常对照组、糖尿病模型组、3种不同剂量葛根素治疗组。糖尿病大鼠模型建立后,葛根素治疗组分别给予葛根素注射液40mg/kg.d、80mg/kg.d、160mg/kg.d腹腔注射,正常对照组、糖尿病模型组腹腔注射相应量的生理盐水,实验第8周末检测空腹血糖(FBG)、尿白蛋白排泄率(UAER)、肌酐(sCr)、尿素氮(BUN);光镜和电镜观察肾皮质形态学改变;免疫组化法和Western blot法检测PDGF-BB定位和半定量表达。结果:与糖尿病模型组比较,3种不同剂量葛根素治疗组FBG、UAER、sCr、BUN明显降低,光镜及电镜下肾脏病理改变有较大改善,肾组织PDGF-BB表达减少。结论:葛根素对糖尿病大鼠肾脏有保护作用,可能与下调肾组织PDGF-BB表达有关。
Objective:To observe the influence on the expression of PDGF-BB of puerarin on diabetic rat's kidney.Methods:SD rats were randomly assigned to normal control group;diabetic model group and three different puerarin dose groups.After the diabetic rat models were founded,three different puerarin dose groups were treated respectively with puerarin 40mg/kg·d,80mg/kg·d,160mg/kg·d to peritoneal inject,normal control group and diabetic model group were treated with corresponding sodium chloride.The FBG,UAER,Scr and BUN were tested at the eighth weekend.Observe the morphologicai change of renal cortex by light microscope and electron microscope.Detect the expression of PDGF-BB by immunohistochemical method and Western blot.Results:Compared with diabetic model group,The FBG,UAER,Scr,BUN in three different puerarin dose groups were decreased markedly(P〈 0.01 or P〈0.05).kidney pathologic changes of diabetic rats in puerarin treatment groups were improved.The expression of PDGF-BB in kidney tissue were decreased significantly(P〈0.01).Conclusion:Puerarin can reduce urinary albumin excretion,improve glycometabolism,renal function and renal constitution of diabetic rat,it play a protective role to diabetic rat kidney which may be concerned with reducing the expression of PDGF-BB.
