摘要
[目的]对水稻农杆菌转化体系中共培养过程进行优化。[方法]以水稻品种日本晴作为材料,对2种不同的共培养方法,即将1层无菌滤纸铺于固体共培养基表面和用0.5 mLAAM培养液浸湿的无菌滤纸的方法进行比较。[结果]无菌滤纸用0.5 mL AAM培养液浸湿后铺于培养皿,后直接将浸染后的愈伤组织在其上进行共培养,效果好且染菌少。[结论]利用0.5 mL AAM浸湿的滤纸培养既简化操作又有利于愈伤组织生长。
[Objective] The co-culture of Agrobacterium-mediated transformation in Oryza sativa was optimized. [Method] With Oryza sativa L.japonica. cv. Nipponbare as the tested material, took two different co-culture methods were set up to study the effect of 0.5 mL AAM on co-culture. [Result] The effect of filter paper soaked with 0.5 mL AAM was good and infected bacteria less. [Conclusion] The filter paper soaked with 0.5 mL AAM was beneficial to the callus formation.
