Kisspeptins在孕早期血浆和胎盘组织中的表达及其意义

摘要 目的:研究kisspeptins在孕早期血浆和胎盘组织中的表达,拟探讨其来源及其在妊娠中意义方法:采用酶联免疫吸附法(ELISA),检测40例孕早期妇女和35例正常未妊娠妇女血浆kisspeptins的表达水平,且分析其与hCG的相关性;采用免疫组织化学方法进行其组织学定位。结果:(1)kisspeptins在妊娠组(982.46±239.41 pmol/L)的表达水平明显高于非妊娠组(3.57±0.37 pmol/L,P<0.001)(2)在妊娠组,血浆中kisspeptins水平与hCG表达水平具有明显的正相关(r=0.84,P=0.015)。(3)免疫组织化学研究显示,kisspeptins强烈表达于子宫蜕膜细胞和绒毛组织的合体滋养层细胞,在细胞滋养层细胞有比较弱的表达,而在绒毛外滋养层细胞却没有表达。结论:在妊娠早期kisspeptins表达水平明显的升高。 Objective:To study the expression levels of kisspeptins in the plasma and placenta tissue,and investigate its origin and significance in early pregnancy.Methods:Using enzyme-linked immunosorbent assay(ELISA),plasma kisspeptins were detected and analyzed its correlation with human chorionic gonadotropin(hCG) in 40 eases of pregnant women and 35 nornial non-pregnant women.Immunohistochemistry revealed histological localization of kisspeptin.Results: (1)Pregnant women had significantly higher plasma kisspeptin levels(982.46±239.41 pmol/L) than nonpregnant women(3.57±0.37pmol/L)(P0.001).(2)Plasma levels of kisspeptins in pregnant women has a signifi- cant positive correlation with hCG(r=0.84,P=0.015).(3) Immunohistocheniistry demonstrated that kisspeptins were strongly expressed in decidual cells and the syncytiotrophoblast,and weakly expressed in cytotrophoblast cells of chorionic villi,but not in extravillous trophoblasts.Conclusion:The expression levels of kisspeptins were significantly increased in early pregnancy,which may come from the decidual and placental tissue,and expression level of kisspeptins in the plasma could be used as a complementary marker for pregnancy.
