摘要
目的检测Toll样受体4(TLR4)在非肥胖型糖尿病(NOD)鼠胰腺的表达及发病过程中的变化,并探讨其临床意义。方法利用免疫荧光定位TLR4在胰腺中的表达,采用逆转录一聚合酶链反应(RT—PCR)及Western blot等方法分别从mRNA和蛋白水平探讨其在胰腺中的表达及发病过程中的变化。结果几乎所有的胰岛p细胞表面均表达TLR4。随着1型糖尿病的进展,胰腺TLR4蛋白相对表达量由1.01±0.01上升到3.68±0.10,组间差异有统计学意义(P〈0.01);mRNA相对表达量由1.00±0.02上升到4.76±0.17,组间差异有统计学意义(P〈0.01)。结论随着1型糖尿病的进展,TLR4表达明显增加,其可能通过免疫机制参与了疾病的发生、发展过程。
Objective To detect the expression of Toll-hke receptor 4 (TLR4) in pancreas of non-obesediabetic (NOD) mice and the change in the pathogenesis in order to explore their clinical significance. Methods Immunofluoreseence was used to detect the location of TLR4 in pancreas, reverse tran-scription-polymerase chain reaction (RT-PCR) and Western blotting were employed to examine its mRNA and protein expression changes in the pathogenesis of type 1 diabetes separately. Results TLR4 was expressed on almost all the surface of 13 islet cells. With the development of the type 1 diabetes, the relative expression of TLR4 was elevated from ( 1.01±0.01 ) to (3.68 ±0. 10), P 〈0. 01; the relative expression of TLR4 mRNA was elevated from ( 1.00±0. 02) to (4. 76±0. 17), P 〈0. 01. Conclusion The expression of TLR4 is significantly increased with the progress of type 1 diabetes, suggesting that TLR4 may participate in the occurrence and development of type 1 diabetes by immune mechanisms.
