摘要
为了探索龙眼核中黄酮类物质的提取工艺,根据Response surface methodology(RSM)中的Box-Behnke中心组合试验设计原理,建立了3因素3水平的响应面分析方法,分别考察了提取温度、溶剂浓度、提取时间对龙眼核黄酮提取的影响,并建立了黄酮提取与各影响因素的多元二次回归数学模型。通过响应面分析法得出龙眼核黄酮最佳提取工艺参数为:提取温度71℃,提取溶剂甲醇的浓度为50%,提取时间为70 min,按照此工艺提取黄酮,测得黄酮得率为1.6388%,与模型预测值1.6164%相符合。
Based on the Box-Behnke centre combination experimental design principle of Response Surface Methodology(RSM),RSM with three factors and three levels was established to investigate the effects of extraction temperature,solvent concentration and extraction time on the extraction of flavonoids from longan(Dimocarpus longan Lour.) seed,and the quadratic polynomial mathematical model describing the relationship between flavonoids extraction and all influential factors was set up.Through RSM,the optimal conditions of extracting flavonoids were obtained as follows: extraction temperature 71 ℃,extracting solvent methanol concentration 50% and extraction time 70 minutes.Under these conditions,the experimental gain rate of flavanoids was 1.6388%,which was basically coincident with the predicted value(1.6164%) of the model.
