摘要
目的探讨细胞外基质金属蛋白酶诱导因子(EMM-PRIN)和基质金属蛋白酶7(MMP-7)在口腔鳞状细胞癌(OSCC)中表达及其临床意义。方法应用免疫组化SP法检测62例手术切除中的OSCC组织及11例正常组织中EMMPRIN和MMP-7的表达水平,并采用RT-PCR检测其中18例OSCC组织及5例正常组织中MMP-7 mRNA的表达。结果 EMMPRIN在OSCC组织中阳性率高于其在正常组织中的阳性率(P<0.05);MMP-7在OSCC组织中阳性表达率高于正常组织(P<0.05);MMP-7 mRNA在OSCC组织中表达高于正常组织(P<0.05)。EMMPRIN表达与肿瘤的大小、临床分期及淋巴结转移密切相关,MMP-7表达则与肿瘤的临床分期和淋巴结转移相关,两者的表达均与患者性别和年龄无关。相关性分析显示EMMPRIN和MMP-7之间呈正相关(r=0.801)。结论 EMMPRIN和MMP-7异常表达与OSCC的生物学行为密切相关,EMMPEIN可能诱导产生MMP-7,进一步导致OSCC的浸润及转移。
Objective To investigate the expression and significance of EMMPRIN and MMP-7 in oral squamous cell carcinoma(OSCC). Methods The expressions of EMMPRIN and MMP-7 were examined by SP immunohistochemical method stain(IHC) in 62 cases of OSCC and 11 cases of normal tissues. The MMP-7 mRNA expression was detected by reverse transcription-polymerase chain reaction (RT-PCR) in 18 cases of OSCC and 5 cases of normal tissues. Results The positive ratio of EMMPRIN in OSCC was significantly higher than that in normal tissues (P 〈 0.05). The positive ratio of MMP-7 in OSCC was higher than that in normal tissues (P 〈 0. 05 ). The MMP-7 mRNA expression in OSCC was higher than that in normal tissues ( P 〈 0.05 ). The EMMPRIN expression was significantly associated with tumor diameter, clinical stage and the lymph node in the samples. The MMP-7 expression was significantly related with clinical stage and the lymph node, and neither of them was correlated with sex, age. EMMPRIN level was positively related to MMP-7 level( r = 0. 801 ). Conclusion EMMPRIN and MMP-7 play an important role in the genesis, development,invasion and metastasis of OSCC.
