新疆地区汉族和哈萨克族心房纤颤患者血管紧张素转换酶基因I/D多态性研究 被引量:5

Polymorphism of ACE Gene I/D of Atrial Fibrillation Patients in Xinjiang Kazakh and Han Nationality

摘要 目的探讨血管紧张素转换酶(ACE)基因I/D与新疆哈萨克族、汉族心房纤颤(AF)之间的关系。方法以哈萨克族、汉族原发性AF患者各100例作为AF组,哈萨克族、汉族健康者各100例作为对照组,提取基因组DNA,采用聚合酶链反应检测ACE基因的I/D多态性。结果哈萨克族AF组与对照组ACE I/D多态位点基因型频率和等位基因频率比较,差异有统计学意义(P<0.05),汉族AF组与对照组ACE I/D多态位点基因型频率和等位基因频率比较,差异有统计学意义(P<0.05),ACE基因I/D多态性位点在汉族和哈萨克族AF组比较,差异有统计学意义(P<0.05)。男性AF组与女性AF组ACE I/D多态位点基因型频率和等位基因频率比较,差异无统计学意义(P>0.05)。结论 ACE基因I/D与新疆地区汉族、哈萨克相关,ACE基因D型等位基因可能是AF的危险因素,ACE基因I/D多态位点在汉族与哈萨克族AF患者分布中存在民族差异。 Objective To discuss the association between ACE gene I/D polymorphism in Xin jiang Kazaks and Hans.Methods Genetic DNA was extracted from 200 primary AF patients including 100 patients from Kazakh and 100 from Han Ethnic group and 200 normal controls including 100 from Kazakh and 100 from Han Ethnic group.The I/D polymorphism of ACE was detected by polymerase chain reaction(PCR),and the ACE gene I/D polymorphism of Chymase gene was detected by PCR restriction fragment length polymorphism(RFLP).Results There was significant difference between Kazakh AF group and Kazakh control group in ACE gene I/D polymorphism genotype frequency and allele frequency(P0.05).There was also significant difference between Han AF group and Han control group in ACE gene I/D polymorphism genotype frequency and allele frequency(P0.05).There was significant difference between Kazakh Ethnic and Han Ethnic in ACE gene I/D polymorphic loci(P0.05).There was no significant difference between male AF and female AF in ACE gene I/D polymorphism genotype frequency and allele frequency(P0.05).Conclusion The I/D polymorphism of ACE gene is associated with the occurrence of AF in Kazakh and Han Ethnic AF population of Xinjiang.Genotype D might be one of the genetic risk factors for AF.There is national difference between Kazakh Ethnic and Han Ethnic in ACE I/D polymorphisic loci.
