香菇多糖对酒精性肝损伤小鼠预防性肝保护作用的研究 被引量:22

摘要 目的观察香菇多糖对小鼠急性酒精性肝损伤的保护作用。方法将昆明种小鼠分为空白对照组、病理模型组、香菇多糖高、中、低剂量保护组。对照组用生理盐水灌胃,病理模型组小鼠用56℃红星二锅头灌胃,造成急性酒精性肝损伤模型。香菇多糖各保护组,先用高、中、低剂量的香菇多糖处理,3 h后分别用56℃红星二锅头(12 mL/kg)灌胃。小鼠饲养10 d末次灌胃禁食不禁水16 h后,眼球取血分离血清,测定ALT和AST值;然后处死小鼠,立即取出肝脏制备肝匀浆,按试剂盒说明书要求检测肝匀浆中SOD、GSH-Px的活性及MDA、GSH的含量。结果香菇多糖能降低ALT、AST活性和AST/ALT比值,增加SOD、GSH-Px的活性,提高GSH含量,降低MDA的含量。结论香菇多糖可能对酒精性肝损伤有保护作用。 【Objective】 To investigate the protective effects of lentin on alcoholic liver injury in mice.【Methods】 Kunming mice were divided into 5 groups: control group,the mice were perfused with chloride isotonicus,pathology model group,the mice were perfused with the 56℃ red star white liquor 10 days to create the acute alcoholic liver injury model continuously,low-dosage lentinan group,intermidiate-dosage lentinan group and high-dosage lentinan group,The mice were perfused with different dosage’s lentinan,3 hours later,the 56℃ red star white liquor were perfused.10 days later,after the latest perfusion,the mice should be keep fasting but not be restained water 16 hours.Then mice were weighed and drew blood from eyeball,the levels of serum ALT and AST were determined.Liver homogenate were prepared to detect MDA and GSH content,SOD and GSH-Px activity.【Results】 The lentinan can reduce the serum levels of ALT and AST,and depress the ratio of AST/ALT,increase SOD and GSH-Px activity,raise GSH content and reduce MDA content.【Conclusion】 The lentinan maybe has protective function on the alcoholic liver injury.
