摘要
目的观察香菇多糖对小鼠急性酒精性肝损伤的保护作用。方法将昆明种小鼠分为对照组、模型组、香菇多糖高、中、低剂量保护组。模型组和香菇多糖各保护组,用56℃红星二锅头连续10 d灌胃小鼠,造成急性酒精性肝损伤模型。保护组用不同剂量的香菇多糖灌胃处理;末次灌胃禁食不禁水16 h后,称取小鼠体重,眼球取血分离血清,测定AST;然后处死小鼠立即取出肝脏制备肝匀浆,按试剂盒要求检测肝匀浆中NOS和iNOS的活性及MDA、NO、TNF-α的含量。结果与对照组比较,酒精性肝损伤模型组小鼠血清AST和肝脏NOS和iNOS的活性升高,MDA、NO及TNF-α的含量增加(P<0.01)。香菇多糖各保护组与模型型组比较,小鼠血清AST和肝脏NOS和iNOS的活性降低,MDA、NO及TNF-α的含量降低(P<0.05-0.01)。结论香菇多糖能够对抗和清除自由基,降低小鼠血清AST和肝脏MDA、NO及TNF-α含量,对酒精性肝损伤有保护作用。
Objective To investigate the protective effect of lentinan on the alcoholic liver injury mice.Methods Kunming mice were divided into control group,model group,low,moderate and high dose lentinan groups.The acute alcoholic liver injury model was induced by gavage with 56 ℃ alcohol(Red Star Erguotou) for 10 d.The mice were gavaged with different doses of lentinan in lentinan groups.Mice were weighed at 16 h after the last gavage time,and serum AST activity was determined.Then the mice were killed,the activities of NOS,iNOS and the contents of MDA,NO,TNF-α were detected in liver homogenate.Results Compared with control group,the activities of serum AST and liver NOS,iNOS in model group increased significantly,and the contents of MDA,NO and TNF-α also increased significantly(P〈0.01).Compared with model group,the activities of serum AST and liver NOS,iNOS reduced in lentinan groups,and MDA,NO,TNF-α contents also decreased(P〈0.05-0.01).Conclusion Lentinan can protect from alcoholic liver injury in mice by fighting against and scavenging free radicals,reducing serum AST,decreasing the activities of NOS,iNOS and contents of MDA,NO and TNF-α in liver.
