TCF7L2基因rs7903146多态性与2型糖尿病的相关性研究 被引量:6

摘要 目的探讨转录因子7样2(TCF7L2)基因rs7903146多态性与中国随州地区汉族人群2型糖尿病的相关性。方法选取已经确诊为2型糖尿病患者458例,其中男232例,女226例;空腹血糖<5.6 mmol/L,无其他系统性疾病的表现健康个体186名,其中男91名,女95名。测量研究对象的一般临床指标,糖代谢和脂代谢相关指标。应用聚合酶链反应(PCR)TaqManTM荧光探针技术进行等位基因分型,对2组共642例研究对象的TCF7L2基因rs7903146多态性进行分析。结果 TCF7L2基因rs7903146多态性符合Hardy-Weinberg平衡定律,TCF7L2基因rs7903146的C/C、C/T、T/T 3种基因型在2型糖尿病组中的频率分别为92.2%、7.6%和0.2%,在对照组的频率为92.5%、7.5%和0%,χ2检验发现,2组间差异无统计学意义。T等位基因频率在糖尿病组和对照组分别为4.0%、3.8%,经χ2检验差异无统计学意义。结论 TCF7L2 rs7903146多态性不是湖北随州地区的汉族人群中2型糖尿病的主要致病基因位点。 Objective To investigate the correlationship of transcription factor 7-like 2 (TCFTL2) gene rs7903146 polymorphism and Chinese Han population in Suizhou district with type 2 diabetes meflitus. Methods A total of 458 patients with type 2 diabetes meliitus (232 males and 226 females) had been diagnosed, and 186 healthy subjects (91 males and 95 females) with fasting blood glucose 〈 5.6 mmol/L and without other systemic diseases were enrolled. The clinical indicators and glucose metabolism and lipid metabolism related indicators were measured. The polymerase chain reaction (PCR) TaqManTM fluorescence probe technique was used for allelie typing. The TCFTL2 gene rs7903146 polymorphism was analyzed. Results TCFTL2 gene rs7903146 polymorphism was consistent with Hardy-Weinberg equilibrium law, and the frequencies of TCF?L2 gene rs7903146 C/C, C/T and T/T genotypes in type 2 diabetes mellitus group were 92.2% , 7.6% and 0.2% , and the frequencies in the control group were 92.5% , 7.5% and 0. No statistical difference between the 2 groups was found by ~2 test. T allele frequencies in the type 2 diabetes mellitus group and control group were 4.0% and 3.8% , without statistical significance. Conclusions TCF7L2 gene rs7903146 polymorphism is not the main pathogenic gene among Chinese Han population in Suizhou district with type 2 diabetes mellitus.
