摘要
目的研究毛橘红总黄酮对酒精性肝损伤大鼠肝细胞凋亡的影响。方法将大鼠随机分为空白对照组、模型组、五子衍宗丸组、毛橘红总黄酮高、中、低剂量组,采用白酒灌胃造成酒精性肝损伤模型,造模同时进行药物治疗,4周后处死动物,于肝小叶切取肝组织制成肝细胞悬液,利用流式细胞仪检测细胞凋亡率。结果与空白对照组比较,模型组细胞凋亡率显著升高(P<0.01),与模型组比较,高剂量组肝细胞的凋亡率有明显下降(P<0.01)。结论毛橘红总黄酮能够抑制肝细胞凋亡,对酒精性肝损伤有保护作用。
Objective To study the effects of total flavonoids of Exocarpium Citri Grandis on hepatocyte apoptosis in rats with alcoholic hepatic injury.Methods Animal model of alcoholic liver damage in rats was established by intragastric administration.Male rats were randomly divided into six groups,including the negative group,the model group,the positive group,and three treatment groups with different dosages of total flavonoids of Exocarpium Citri Grandis.After four weeks,liver tissues were extracted and made single-cell suspensions.The rate of hepatocyte apoptosis was detected by flow cytometry.Results Compared with the negative group,apoptotic rate was increased in the model group(P〈0.01),but the increased apoptotic rate was reduced when treatment with high-dosage of total flavonoids of Exocarpium Citri Grandis.Conclusion Total flavonoids in Exocarpium Citri Grandis can inhibit hepatocyte apoptosis and exert protective effect against alcoholic hepatic injury.
