摘要
目的 本文旨在研究三苯氧胺在膀胱癌T -2 4细胞株中可能有的化疗增敏作用。材料与方法 应用MTT方法 ,以膀胱肿瘤T-2 4细胞株为研究对象 ,比较单独应用化疗药物 (ADM、MMC、Thiotepa、VCR)与化疗药物合并应用三苯氧胺 (Tamoxifen 5 μM ,10 μM )对细胞生长抑制率效应 ,通过化疗药物的量—效关系曲线 ,观察三苯氧胺是否有特定的化疗增敏作用。结果 三苯氧胺在浓度为 5μM、10 μM时 ,能分别使ADM和VCR的IC50 下降 1 95、7 0 6倍和 6 94、14 5 5倍 ,差异有显著性 (P <0 0 5 ) ;而三苯氧胺在 5 μM、10 μM的浓度下 ,对MMC、Thiotepa的IC50 的影响 ,无统计学意义。结论 在体外试验中 ,三苯氧胺在 5 μM和 10 μM的浓度下能显著性地促进膀胱肿瘤T -2
Objective The aim of this study is to evaluate the possible effects of Tamoxifen on chemosensitization of T-24 bladder carcinoma cell line.Meterials and methods In vitro,chemosensitivity tests were done with T-24 bladder carcinoma cell line and 4 cytotoxic agents (Adriamycin,ADM;Mitomycin C,MMC;Thiotepa,Thiot; Vincristine,VCR) in the presence or absence of graded concentration of Tamoxifen (5 μM,10 μM)。Growth inhibition tests were performed with Tetrazolium (MTT) assay.Through the curves of growth inhibition of T-24 treated with grade concentration of cytotoxic agents,the chemosensitization of Tamoxifen was observed.Results Tamoxifen at the concentration of 5 μM and 10 μM increased significantly the chemosensitivity of ADM by 1.95 to 7.06 fold and the chemosensitivity of VCR by 6.94 to 14.55 fold,respectively.The chemosensitivity of ADM and VCR could be significantly protentiated by Tamoxifen at the concentration of 5 μM and 10 μM ( P <0 05).But Tamoxifen at 5 μM and 10 μM could not increase the chemosensitivity of MMC and Thiot,significantly.Conclusions In vitro,Tamoxifen at the concentration of 5 μM and 10 μM can significantly enhance chemosensitivity of ADM and VCR to T-24 bladder cancer cell line.For less side effects of Tamoxifent at such concentration,it might be a safe,potential chemosensitizer in systemic treatment or intravesical instillation to human bladder cancers.
