摘要 目的:研究蓝萼甲素的抗血栓作用。方法:考察蓝萼甲素对SD大鼠的血小板聚集性、血瘀模型大鼠血浆中TXB2、6-Keto-PGF1α以及NO和ET含量的影响,通过大鼠动-静脉旁路血栓形成实验、ADP-肾上腺素诱发小鼠体内血栓形成实验、急性血瘀模型大鼠血液流变学实验、大鼠离体动脉条的舒张实验,研究蓝萼甲素的抗血栓作用,并初步探讨其机制。结果 :蓝萼甲素浓度依赖性降低ADP以及凝血酶所诱导的血小板聚集;降低血瘀模型大鼠血浆中TXB2含量,显著升高血浆中6-KetoPGF1α含量,升高血浆中NO的含量,降低血浆中ET含量;蓝萼甲素对大鼠动-静脉旁路血栓形成有抑制作用,对ADP-肾上腺素混合溶液诱导的小鼠血栓性偏瘫形成有保护作用;显著降低急性血瘀模型大鼠的全血及血浆黏度,蓝萼甲素依赖内皮细胞发挥舒张血管作用,而且与NO合成通路密切相关。结论:蓝萼甲素通过抑制血小板聚集,抑制血栓形成,降低全血及血浆黏度,舒张血管,从而发挥抗血栓作用。 Objective:The study was aimed to investigate the antithrombotic activity of glaucocalyxin A.Methods:It studied the effect of glaucocalyxin A to the levels of TXB2 and 6-Keto-PGF1α,NO and ET in plasma of model rats with blood stasis in vitro.The influence of glaucocalyxin A on thrombosis was observed by rats artery-vein bypass thrombosis experiments and mice tail vein thrombosis experiments,the effects of glaucocalyxin A on blood rheology of blood stasis model animals by monitoring blood viscosity and plasm viscosity of model rats with blood stasis,the effects of glaucocalyxin A on rat thoracic aorta were studied.Results:Glaucocalyxin A can inhibit the platelet aggregation induced by ADP and thrombin.In addition,glaucocalyxin A can obviously increase the levels of 6-KetoPGF1α and NO of ice water-adrenalin blood stasis model rats,and obviously decrease the level of ET.Glaucocalyxin A can decrease thrombosis in the rats artery-vein bypass thrombosis experiments and inhibit thrombosis induced by the ADP-epinephrine.Glaucocalyxin A can reduce blood and plasm viscosity.Glaucocalyxin A has vasodilative effects depending on endothelial cells,and NO synthase inhibitor L-NAME can significantly weaken the vasodilative effects of glaucocalyxin A,which effects are mainly achieved through the NO-sGC-cGMP pathway.Conclusion:Glaucocalyxin A has obvious antithrombotic activity,and it can reduce platelet aggregation rate,increase levels of 6-KetoPGF1α and NO as well as decrease the level of ET,improve blood rheology,diastolic vessels,effectively lighten artery-vein bypass thrombosis,and improve pulmonary embolism of mice.
